Pharmacokinetics in children

In adolescents between 12 and under 18 years of age, a small difference was observed in half-life (n=5, 72.8 ± 16,5 h) compared to adults (n=16, 76.9 ± 26,1 h). 
Clearance was nearly identical in both age groups and was 0.68 ± 0.18 mL/h/kg and 0.66 ± 0.21 mL/h/kg, respectively [SmPC Fibryga].

The following pharmacokinetic parameters were determined in children under 12 years of age with congenital fibrinogen deficiency (n=13, dose: 70 mg/kg i.v. single dose) [SmPC Fibryga]:

Treatment of bleeding
  • Intravenous
    • 1 month up to 18 years
      • Depending on the clinical necessity, usually 20 - 30 mg/kg/dose, as required.
      • Directions for administration:

        Administer slowly, max. 5 ml/min.

Prophylaxis in case of congenital hypo-dysia or afibrinogenaemia and known bleeding tendency
  • Intravenous
    • 1 month up to 12 years
      • Dose (mg/kg) = [Target level (g/L) – measured level (g/L] / 0,014 (g/L per mg/kg bodyweight)

    • Term neonate
