Dosages

GTP cyclohydrolase I deficiency (GTPCH); 6 pyruvoyl tetrahydropterin synthase deficiency (PTPS); dihydropteridine reductase deficiency (DHPR)
  • Oral
    • Term neonate
      • Levodopa:carbidopa = 10:1
        Dose according to the levodopa:
        1 - 3
        mg/kg/day in 3 - 6 doses. Increase gradually over a few days/weeks in steps of no more than 1 mg/kg..
      • Treatment by or after consultations with a paediatric specialist (metabolic disorders) who has experience using levodopa-carbidopa for this indication.

    • 1 month up to 2 years
      • Levodopa:carbidopa = 10:1
        Dose according to the levodopa: 4 - 7
        mg/kg/day in 3 - 6 doses. Increase gradually over a few days/weeks in steps of no more than 1 mg/kg per day/week.
      • Treatment by or after consultations with a paediatric specialist (metabolic disorders) who has experience using levodopa-carbidopa for this indication.

    • ≥ 2 years
      • Levodopa:carbidopa = 10:1
        Dose according to the levodopa 8 - 15
        mg/kg/day in 3 - 6 doses. Increase gradually over a few days/weeks in steps of no more than 1 mg/kg per day/week..
      • Treatment by or after consultations with a paediatric specialist (metabolic disorders) who has experience using levodopa-carbidopa for this indication.

Tyrosine hydroxylase deficiency
  • Oral
    • 0 years up to 18 years
      • Dose according to the levodopa: 1 - 10 mg/kg/day in 2 - 6 doses. . Increase gradually over a few days/weeks in steps of no more than 1 mg/kg..
      • Use the 25% product (levodopa:carbidopa 4:1) when the total daily dose of levodopa is less than 400 mg, use the 10% product (levodopa:carbidopa 10:1) otherwise.
